Bekannt ist sie vor allem für ihre Hauptrolle als Blair Waldorf in der Teenager-Drama-Serie Gossip Girl (2007-12). Sie war auch in dem Landdrama-Film Country Strong 2010, dem Thriller The Roommate 2011, der Romantikkomödie Monte Carlo 2011 und den Komödien The Oranges (2011) und That's My Boy (2012) zu sehen. Meester gab ihr Broadway-Debüt 2014 in Of Mice and Men.
